CRISPR-Cas9

CRISPR/Cas9 edits genes by precisely cutting DNA and then letting natural DNA repair processes to take over. The system consists of two parts: the Cas9 enzyme and a guide RNA.

Neural Organoids

Neural organoids are hPSC-derived 3D in vitro culture systems that recapitulate the organizational processes of the developing human brain. We make use of unguided cerebral organoids and guided forebrain and choroid plexus organoids.

Imaging

To investigate the morphology, proliferation, and cellular architecture of our 2D & 3D models, we employ immunohistochemistry (IHC), fluorescence-lifetime imaging (FLIM), and live-cell monitoring. 

Transcriptomics

RNA-seq allows detection of qualitative and quantitative changes in RNA expression across our models. These changes can be determined either in bulk or at the single-cell level. For integrative analyses, we combine this with our epigenomic toolkit.

Chromatin confirmation capture

Chromatin conformation capture techniques are a set of molecular biology methods used to analyze the spatial organization of chromatin in a cell. In the FunGen lab, we employ UMI-4C, Hi-C, and Capture-C.

Epigenomics

Epigenomics is used to explore and identify candidate cis-regulatory elements. Our epigenomics toolkit consists of ATAC-seq, CUT&RUN, nanoCUT&Tag to determine open chromatin, histon modifications, and transcription factor binding sites.
